We have an application form on our site for customers to open a wholesale account. Over the last year we've set up well over 100 accounts but many have never been used. Is there any way of checking when accounts were set up (perhaps a record in the database) so I can write to the oldest customers to check if they're ever likely to use the accounts?

You can use the marketing / mailing list to get a list of customers that have never ordered.
Go to marketing / mailing lists, click on the export customer button, put in a date range that will include the time period you have been adding customers for and then click on preview to see a list of customers that have not placed orders in that time period or click on export to create a csv file with that same list.
