    Customising a Specific Design Theme

The general advice is to customise Clean Layouts. However none of those listed resemble BusinessCSS ie a fixed width centred theme.

Are there real drawbacks to customising a Specific Design?

business css is fine to customise, as are the clean layouts, once you start customising you can fix width, centre etc etc, so the initial base theme is not too important

Always choose a css based theme and rip it apart!

As Jo says you can customise any theme - deciding on which to customise depends on which is closest to the overall design and feel you wish to end up with... it is often simplest to use one of the clean CSS themes as there is less "clutter" to remove and just the basic elements are there - but if a specific theme such as smart or business is almost the design you are looking for then it may be quickest to start there and start the changes
